A new kinetic model proposed for enzymatic hydrolysis of lactose by a -galactosidase from Kluyveromyces fragilis

چکیده
We study the enzymatic hydrolysis of lactose by a commercial enzyme from a selected strain of Kluyveromyces fragilis. The variables analyzed were: temperature (25–40 ◦C), enzyme concentration (0.1–3.0 g l−1), lactose concentration (0.0278–0.208 M), and initial galactose concentration (0.0347 M). On the basis of the data analyzed, both published and in the present work, we propose a Michaelis–Menten kinetic model with inhibition by the product (galactose), which reveals that the substrate (lactose) and the product (galactose) present similar affinity for the active site of the enzyme. © 2002 Elsevier Science Inc. Bioaffinity Based Immobilization of Almond (Amygdalus communis) b-galactosidase on Con A-layered Calcium Alginate-cellulose Beads: Its Application in Lactose Hydrolysis in Batch and Continuous Mode
In this study, immobilization of partially purified almond (Amygdalus communis) β-galactosidase on Con A layered calcium alginate-cellulose beads was investigated. Immobilized β-galactosidase retained 72% of theinitial activity after crosslinking by glutaraldehyde. Both soluble and immobilized enzyme exhibited the samepH and temperature optima at pH 5.5 and 50ºC, respectively. Howev...
متن کاملLactose Hydrolysis in Milk and Dairy Whey Using Microbial β-Galactosidases
This work aimed at evaluating the influence of enzyme concentration, temperature, and reaction time in the lactose hydrolysis process in milk, cheese whey, and whey permeate, using two commercial β-galactosidases of microbial origins. We used Aspergillus oryzae (at temperatures of 10 and 55°C) and Kluyveromyces lactis (at temperatures of 10 and 37°C) β-galactosidases, both in 3, 6, and 9 U/mL c...
متن کاملInfluence of the hollow-fibre membrane on the stability of -galactosidase and on lactose hydrolysis Kinetic models including adsorption of the enzyme onto the membrane
An adsorption-kinetic model has been proposed to explain -galactosidase stability in a recirculation hollow-fibre bioreactor at different temperature. The kinetic constants of the model and its dependence on temperature have been evaluated. In addition, a study has been made of the influence of lactose and enzyme concentration on lactose hydrolysis with hollow-fibre modules m s a m a ©
متن کاملKinetics and Regulation Studies of the Production of -Galactosidase from Kluyveromyces marxianus Grown on Different Substrates
Lactose-intolerance is manifested in 50 % of the world’s population. This can be remediated by removing lactose from the diet or converting it into glucose and galactose with -galactosidase (EC 3.2.1.23). In this work, batch production of this enzyme in the presence of lactose, galactose, cellobiose, xylose, arabinose, sucrose and glucose was investigated using Kluyveromyces marxianus in shake ...
متن کاملGalactooligosaccharides formation during enzymatic hydrolysis of lactose: towards a prebiotic-enriched milk.
The formation of galactooligosaccharides (GOS) in skim milk during treatment with several commercial β-galactosidases (Bacillus circulans, Kluyveromyces lactis and Aspergillus oryzae) was analysed in detail, at 4 and 40°C. The maximum GOS concentration was obtained at a lactose conversion of approximately 40-50% with B. circulans and A. oryzae β-galactosidases, and at 95% lactose depletion for ...
